Die Universität Witten/Herdecke lädt ein zum ZBAF-Kolloquium mit Dr. Doris Höglinger (Junior group leader, Biochemistry Center (BZH), Heidelberg University). 

“Lipid transport across contact sites of endocytic organelles with the ER” Late endocytic organelles such as late endosomes and lysosomes are indispensable for functional lipid homeostasis. Hydrolytic enzymes contribute to breakdown of complex lipids to simpler building blocks followed by controlled export of these reaction products for reuse in biosynthetic organelles. This export of lipids from late endocytic organelles is especially important for sphingolipid and cholesterol homeostasis. Organelle contact sites, microdomains of close membrane apposition, are gaining attention as important platforms for non-vesicular, inter-organellar communication. We introduce a novel role for Niemann-Pick type C protein 1 (NPC1) in tethering contacts between late endocytic organelles and the ER and discuss its impact on lipid homeostasis. Absence or inhibition of NPC1 resulted in reduced contacts of late endosomes and lysosomes with the ER leading to significantly slower lipid transport and metabolism as investigated using functionalized lipid probes. Surprisingly, the cholesterol accumulation characteristic for NPC1-deficient cells could be rescued by artificially tethering late endocytic organelles to the ER. This challenges the notion that the primary function of NPC1 is that of a cholesterol transport protein. Instead, we show that NPC1 interacts with an ER-resident sterol transport Gramd1b in a sterol-responsive manner to provide platforms for lipid export from late endocytic organelles.
